Abstract
Diare merupakan masalah kesehatan dengan angka kejadian
tinggi dan dapat menimbulkan komplikasi serius. Daun nangka
(Artocarpus heterophyllus Lam.) secara empiris digunakan sebagai obat
tradisional antidiare karena mengandung flavonoid, tanin, dan senyawa
fenolik. Variasi konsentrasi pelarut etanol pada proses ekstraksi diduga
memengaruhi kandungan senyawa aktif dan efektivitas farmakologinya.
Penelitian ini bertujuan untuk mengetahui konsentrasi etanol yang
menghasilkan aktivitas antidiare terbaik, kadar fenol total tertinggi, serta
hubungan antara kadar fenol total dengan aktivitas antidiare ekstrak daun
nangka.
Ekstraksi daun nangka dilakukan dengan metode maserasi
menggunakan etanol 50%, 70%, dan 96%. Penetapan kadar fenol total
dilakukan dengan metode Folin–Ciocalteu menggunakan
spektrofotometer UV-Vis. Uji aktivitas antidiare dilakukan secara in vivo
pada mencit jantan yang diinduksi oleum ricini menggunakan metode
transit intestinal. Parameter yang diamati adalah rasio panjang usus yang
dilalui marker norit terhadap panjang usus total. Data dianalisis
menggunakan One-way ANOVA dan uji lanjut Tukey dengan program
SPSS.
Hasil penelitian menunjukkan ekstrak etanol 70% memberikan
aktivitas antidiare terbaik dengan metode transit intestinal (48,12%)
yang sebanding dengan kontrol positif (43,30%), tetapi yang
menghasilkan kadar fenol total tertinggi yaitu ekstrak etanol 96%. Maka,
kadar fenol total tertinggi tidak menghasilkan aktivitas antidiare terbaik,
karena metabolit sekunder yang bertanggung jawab sebagai antidiare
seperti flavonoid dan tanin yang berperan dalam menurunkan motilitas
usus dan sekresi cairan terdapat pada ekstrak etanol dengan pelarut 70%.
Diarrhea is a health problem with a high incidence and can lead
to serious complications. Jackfruit leaves (Artocarpus heterophyllus
Lam.) are empirically used as a traditional antidiarrheal remedy due to
their flavonoid, tannin, and phenolic compounds. Variations in ethanol
solvent concentration during the extraction process are thought to affect
the active compound content and its pharmacological effectiveness. This
study aimed to determine the ethanol concentration that produces the best
antidiarrheal activity, the highest total phenol content, and the
relationship between total phenol content and the antidiarrheal activity
of jackfruit leaf extract.
Jackfruit leaf extraction was carried out using the maceration
method using 50%, 70%, and 96% ethanol. Total phenol content was
determined using the Folin–Ciocalteu method using a UV-Vis
spectrophotometer. Antidiarrheal activity testing was conducted in vivo
in male mice induced by oleum ricini using the intestinal transit method.
The parameter observed was the ratio of the length of the intestine
traversed by the norit marker to the total length of the intestine. Data
were analyzed using one-way ANOVA and Tukey's test using SPSS.
The results showed that 70% ethanol extract provided the best
antidiarrheal activity with the intestinal transit method (48.12%) which
was comparable to the positive control (43.30%), but the one that
produced the highest total phenol content was the 96% ethanol extract.
Therefore, the highest total phenol content did not produce the best
antidiarrheal activity, because secondary metabolites responsible for
antidiarrheal properties such as flavonoids and tannins that play a role in
reducing intestinal motility and fluid secretion are found in the ethanol
extract with 70% solvent.
