Abstract
Diabetes melitus merupakan penyakit metabolik yang ditandai
dengan hiperglikemia. Tanaman obat sebagai terapi alternatif terus
dikembangkan, salah satunya daun jeruk nipis (Citrus aurantifolia)
berpotensi sebagai antihiperglikemik. Penelitian ini bertujuan untuk
mengetahui aktivitas antihiperglikemik fraksi ekstrak etanol daun jeruk
nipis serta menentukan fraksi paling aktif pada tikus putih jantan yang
diinduksi aloksan.
Daun jeruk nipis diekstraksi dengan metode maserasi
menggunakan etanol 96% dan difraksinasi dengan n-heksana, etil
asetat, dan air. Hewan uji dibagi menjadi enam kelompok perlakuan,
yaitu kelompok kontrol negatif, kontrol positif,kelompok ekstrak daun
jeruk nipis (250 mg/kg BB), fraksi n-heksan 6,67 mg/200 g BB tikus,
fraksi etil asetat 6,67 mg/kg BB tikus, dan fraksi air 30 mg/200 g BB
tikus. Perlakuan diberikan selama 28 hari dengan pengukuran kadar
glukosa darah menggunakan glukometer dan dianalisis menggunakan
uji One Way ANOVA dilanjutkan uji Post Hoc Tukey.
Hasil penelitian menunjukkan bahwa semua fraksi memiliki
aktivitas antihiperglikemik yang ditunjukkan oleh penurunan kadar
glukosa darah secara signifikan dibandingkan kontrol negatif serta
fraksi etil asetat merupakan fraksi paling aktif, dimana pada dosis 6,67
mg/200 g bobot badan tikus menunjukkan aktivitas antihiperglikemik
sebanding dengan kontrol positif glibenklamid.
Diabetes mellitus is a metabolic disease characterized by
hyperglycemia. Medicinal plants as alternative therapy continue to be
developed, one of which is lime leaves (Citrus aurantifolia), which are
considered to have antihyperglycemic potential. This study aimed to
determine the antihyperglycemic activity of ethanol extract fractions of
lime leaves and to identify the most active fraction in alloxan-induced
male white rats.
Lime leaves were extracted using the maceration method with
96% ethanol and fractionated with n-hexane, ethyl acetate, water. The
experimental animals were divided into six treatment groups: negative
control, positive control, lime leaf extract group (250 mg/kg BW), n�hexane fraction (6.67 mg/200 g BW), ethyl acetate fraction (6.67
mg/200 g BW), and aqueous fraction (30 mg/200 g BW). Treatments
were administered for 28 days, with blood glucose levels measured
using a glucometer. The data were analyzed using One-Way ANOVA
followed by the Tukey Post Hoc.
The results showed that all fractions exhibited
antihyperglycemic activity, as indicated by a significant reduction in
blood glucose levels compared to the negative control group. The ethyl
acetate fraction was identified as the most active fraction, and at a dose
of 6.67 mg/200 g BW, it demonstrated antihyperglycemic activity
comparable to the positive control glibenclamide.
