Abstract
Kanker payudara merupakan salah satu penyebab utama
kematian pada perempuan, sehingga diperlukan pencarian agen
antikanker baru yang aman. Daun sirih hijau (Piper betle L.) diketahui
mengandung senyawa aktif seperti flavonoid, fenol, dan alkaloid yang
berpotensi sebagai antikanker. Penelitian ini bertujuan untuk
mengetahui aktivitas sitotoksik dan selektivitas fraksi n-heksan, etil
asetat, dan air dari ekstrak etanol daun sirih hijau terhadap sel kanker
payudara T47D dibandingkan dengan sel normal Vero.
Ekstraksi dilakukan dengan metode maserasi, kemudian
difraksinasi dengan metode Ekstraksi Cair-Cair. Uji sitotoksik
dilakukan menggunakan metode MTT, sedangkan selektivitas
ditentukan berdasarkan nilai Indeks Selektivitas (IS). Data dianalisis
menggunakan Microsoft Excel untuk perhitungan persentase viabilitas
sel, kemudian dilakukan analisis regresi linier dan grafik untuk
penentuan nilai IC50 menggunakan GraphPad Prism.
Hasil penelitian menunjukkan bahwa fraksi n-heksan dan fraksi
air tidak memiliki aktivitas sitotoksik terhadap sel kanker payudara
T47D dan sel normal Vero dengan nilai IC50 sebesar >500 µg/mL.
Fraksi etil asetat memiliki aktivitas sitotoksik lemah terhadap sel T47D
dengan nilai IC50 sebesar 357,93 µg/mL, namun juga tidak bersifat
sitotoksik terhadap sel Vero IC50 >500 µg/mL dan memiliki
selektivitas yang rendah dengan nilai Indeks Selektivitas sebesar 1,39.
Breast cancer is one of the leading causes of death among
women, highlighting the need to search for safe anticancer agents.
Green betel leaf (Piper betle L.) is known to contain bioactive
compounds such as flavonoids, phenolics, and alkaloids that have
potential anticancer activity. This study aimed to evaluate the cytotoxic
activity and selectivity of the n-hexane, ethyl acetate, and water
fractions of the ethanol extract of green betel leaf against T47D breast
cancer cells compared with normal Vero cells.
The extraction was carried out using the maceration method,
followed by fractionation using the Liquid-Liquid Extraction method.
Cytotoxicity was assessed using the MTT assay, while selectivity was
determined based on the Selectivity Index (SI) value. The data were
analyzed using Microsoft Excel to calculate the percentage of cell
viability, followed by linear regression analysis and graphing for IC50
determination using GraphPad Prism.
The results of the study showed that the n-hexane and water
fractions exhibited no cytotoxic activity against T47D breast cancer
cells and normal Vero cells, with IC50 values >500 µg/mL. The ethyl
acetate fraction showed weak cytotoxic activity against T47D cells with
an IC50 value of 357,93 µg/mL however, it was also not cytotoxic to
Vero cells IC50 >500 µg/mL and exhibited low selectivity with a
Selectivity Index value of 1,39.
