Abstract
Hiperglikemia diketahui menghambat proses penyembuhan luka
melalui gangguan perfusi jaringan dan peningkatan stres oksidatif.
Serabut kelapa (Cocos nucifera L.) mengandung flavonoid, tanin, dan
saponin yang memiliki aktivitas antioksidan, antibakteri, dan
antidiabetes sehingga berpotensi mempercepat penyembuhan luka.
Penelitian ini bertujuan untuk mengetahui pengaruh variasi konsentrasi
ekstrak etanol serabut kelapa terhadap penyembuhan luka insisi pada
kelinci hiperglikemia yang diinduksi aloksan, serta menentukan
konsentrasi paling efektif untuk diformulasikan dalam sediaan gel.
Penelitian dilakukan menggunakan metode maserasi dengan
etanol 96% untuk memperoleh ekstrak, yang kemudian dalam tiga
konsentrasi, yaitu 1%, 5%, dan 10%, di larutkan menggunakan
dimethyl sulfoxide (DMSO). Ekstrak diuji pada kelinci New Zealand
White yang mencapai kondisi hiperglikemia (≥140 mg/dL). Parameter
diamati meliputi perubahan panjang luka selama proses penyembuhan,
yang dianalisis untuk menentukan konsentrasi paling efektif.
Konsentrasi 10% kemudian diformulasikan menjadi sediaan gel dan
diuji mutu fisiknya, mencakup organoleptik, homogenitas, pH,
viskositas, daya sebar, dan daya lekat.
Hasil penelitian menunjukkan bahwa peningkatan konsentrasi
ekstrak berpengaruh terhadap percepatan penyembuhan luka insisi pada
kelinci hiperglikemia. Konsentrasi 10% memberikan efek paling
optimal dan mendekati kontrol positif. Sediaan gel yang dihasilkan
memenuhi semua persyaratan mutu fisik gel yang baik. Dengan
demikian, gel ekstrak serabut kelapa berpotensi digunakan sebagai
alternatif terapi topikal untuk luka pada kondisi hiperglikemia.
Hyperglycemia is known to inhibit the wound-healing process
by disrupting tissue perfusion and increasing oxidative stress. Coconut
fiber (Cocos nucifera L.) contains flavonoids, tannins, and saponins,
which possess antioxidant, antibacterial, and antidiabetic activities,
thereby offering potential to accelerate wound healing. This study
aimed to determine the effect of varying concentrations of coconut fiber
ethanol extract on the healing of incision wounds in alloxan-induced
hyperglycemic rabbits, as well as to identify the most effective
concentration for gel formulation.
The extract was obtained through maceration using 96% ethanol
and subsequently prepared in three concentrations—1%, 5%, and
10%—dissolved in dimethyl sulfoxide (DMSO). The extracts were
tested on New Zealand White rabbits that reached a hyperglycemic state
(≥140 mg/dL). The primary parameter observed was the change in
wound length during the healing process, which was analyzed to
determine the most effective concentration. The 10% extract was then
formulated into a gel and evaluated for its physical quality, including
organoleptic properties, homogeneity, pH, viscosity, spreadability, and
adhesiveness.
The results showed that increasing extract concentrations
enhanced the rate of incision wound healing in hyperglycemic rabbits.
The 10% concentration demonstrated the most optimal effect and
closely approached the positive control. The resulting gel met all
physical quality requirements for a good topical gel. Therefore, coconut
fiber extract gel has potential as an alternative topical therapy for
wounds under hyperglycemic conditions.
