Abstract
Luka merupakan kerusakan integritas kulit yang sering terjadi
dan memerlukan penanganan khusus untuk mencegah komplikasi. Daun
kenikir (Cosmos caudatus Kunth.) telah lama digunakan dalam
pengobatan tradisional termasuk digunakan dalam penyembuhan luka
karena mengandung senyawa bioaktif seperti flavonoid, alkaloid, tanin,
saponin dan triterpenoid yang dikenal memiliki aktivitas antiinflamasi,
antioksidan, dan antimikroba. Penelitian ini bertujuan untuk melihat
aktivitas ekstrak dan fraksi daun kenikir terhadap penyembuhan luka
sayat pada kelinci New Zealand.
Ekstraksi dilakukan menggunakan etanol, kemudian dilanjutkan
dengan fraksinasi. Uji penyembuhan luka dilakukan melalui pembuatan
luka sayat pada punggung kelinci. Hewan uji dibagi menjadi kelompok
kontrol negatif (vaselin album), kontrol positif (salep MEBO), ekstrak
daun kenikir, fraksi n-heksan, fraksi etil asetat dan fraksi air dengan
konsentrasi setara 15%. Dengan pengamatan selama 14 hari,
berdasarkan pengukuran panjang luka, persentase penutupan luka, serta
pemgamatan makroskopis pada luka.
Hasil penelitian menunjukkan bahwa ekstrak dan fraksi etil asetat
dapat mempercepat penyembuhan luka, dengan fraksi etil asetat paling
efektif yang ditandai dengan penutupan luka 100% pada hari ke-11,
sebanding dengan kontrol positif. Efek ini diduga berkaitan dengan
kemampuan fraksi etil asetat dalam mendukung regenerasi jaringan.
Wounds are a common disruption of skin integrity and require
special treatment to prevent complications. Cosmos caudatus leaves
(Cosmos caudatus Kunth.) have long been used in traditional medicine,
including for wound healing, due to their bioactive compounds such as
flavonoids, alkaloids, tannins, saponins, and triterpenoids, known for
their anti-inflammatory, antioxidant, and antimicrobial activities. This
study aimed to determine the activity of cosmos caudatus leaf extracts
and fractions on wound healing in New Zealand rabbits.
Extraction was performed using ethanol, followed by
fractionation. Wound healing testing was performed by creating
incisions on the rabbits' backs. The animals were divided into negative
control groups (Vaseline album), positive control (MEBO ointment),
cosmos caudatus leaf extract, N-hexane fraction, ethyl acetate fraction,
and water fraction at 15% concentrations. Observations were made over
a 14-day period, based on measurements of wound length, wound closure
percentage, and macroscopic observations.
The results showed that the ethyl acetate extract and fractions
accelerated wound healing, with the most effective ethyl acetate fraction
showing 100% wound closure on day 11, comparable to the positive
control. This effect is thought to be related to the ethyl acetate fraction's
ability to support tissue regeneration.
