Abstract

Tanaman pulutan merupakan salah satu tanaman yang mempunyai efek sebagi obat. Pelarut etanol dapat menarik senyawa-senyawa polar diantaranya glikosida flavonoid, glikosida saponin, fenol dan tanin. Tujuan penelitian ini adalah untuk mengetahui perbandingan variasi konsentrasi etanol 96% dan etanol 70% terhadap kadar fenol total ekstrak daun pulutan (Urena lobata L.). Ekstrak kental daun pulutan didapatkan dengan menggunakan ekstraksi maserasi. Pada konsentrasi ekstrak daun pulutan etanol 96% dan ekstrak etanol 70% dilakukan pengujian secara kualitatif dengan parameter uji reaksi warna, dan uji kromatografi lapis tipis (KLT). Pada pengujian secara kuantitatif ekstrak daun pulutan etanol 96% dan ekstrak daun pulutan etanol 70% dilakukan uji penentapan kadar fenol dengan analisis kalorimetri. Hasil penetapan kadar fenol total pada ekstrak daun pulutan etanol 96% dan 70% masing-masing sebesar 301,4146 mgGAE/gram, dan 220,6188 mgGAE/gram. Nilai kadar fenol total dapat dipengaruhi oleh konsentrasi pelarut. Nilai ini kemungkinan tidak hanya senyawa fenol saja yang terbaca, melainkan juga senyawa seperti flavonoid, tanin, dan vitamin dapat terbaca. Dari hasil tersebut maka dapat disimpulkan bahwa ekstrak etanol 96% dapat lebih optimal dalam menetapkan kadar fenol total dibandingkan dengan ekstrak etanol 70%. The pulutan plant is a plant known for its medicinal properties. Ethanol can extract polar compounds, including flavonoid glycosides, saponin glycosides, phenols, and tannins. The purpose of this study was to determine the ratio of 96% and 70% ethanol concentrations to the total phenol content of pulutan (Urena lobata L.) leaf extract. The concentrated pulutan leaf extract was obtained using maceration extraction. The 96% and 70% ethanol extracts were qualitatively tested using color reaction tests and thin-layer chromatography (TLC) tests. Quantitatively, the 96% and 70% ethanol extracts were tested to determine phenol content using calorimetry. The results of the total phenol content determination in pulutan leaf extracts with 96% and 70% ethanol were 301.4146 mgGAE/gram and 220.6188 mgGAE/gram, respectively. Total phenol content can be influenced by solvent concentration. This value likely includes not only phenol compounds but also compounds such as flavonoids, tannins, and vitamins. From these results, it can be concluded that 96% ethanol extract is more optimal in determining total phenol content compared to 70% ethanol extract.
