Abstract

Fiksasi merupakan tahap awal penting dalam pembuatan sediaan histopatologi, berfungsi untuk mempertahankan struktur jaringan dan mencegah autolisis. Larutan yang paling umum digunakan adalah Neutral Buffer Formalin (NBF) 10%, namun memiliki sifat toksik dan karsinogenik. Oleh karena itu, penelitian ini bertujuan untuk mengetahui gambaran mikroskopis jaringan hati ayam yang difiksasi menggunakan NBF 10% dan madu (Dimocarpus Longan) sebagai alternatif alami, dengan konsentrasi 5%, 15%, dan 100%. Penelitian dilakukan secara eksperimental di Laboratorium Patologi Anatomi RSUD dr. Soeratno Gemolong, menggunakan jaringan hati ayam broiler yang difiksasi selama 24 jam. Fiksasi menggunakan NBF 10% dan madu (Dimocarpus Longan) dengan konsentrasi 5%, 15%, dan 100%. Setelah proses histoteknik dan pewarnaan Hematoksilin-Eosin (HE), dilakukan pengamatan mikroskopis. Hasil penelitian menunjukkan bahwa jaringan hati yang difiksasi dengan NBF 10% memberikan gambaran mikroskopis yang baik dan struktur sel yang utuh. Sementara itu, madu dengan konsentrasi 5% dan 15% menyebabkan jaringan hancur. Pada konsentrasi madu 100%, fiksasi menghasilkan jaringan dengan nekrosis, inti piknotik, dan struktur yang tidak optimal. Kesimpulannya, madu belum mampu menggantikan NBF 10% sebagai fiksatif yang efektif dalam mempertahankan struktur mikroskopis jaringan hati ayam. Madu hanya memberikan hasil kurang baik, terutama pada konsentrasi rendah. Diperlukan penelitian lanjutan untuk menguji alternatif fiksatif alami lainnya. Fixation is a crucial initial step in the preparation of histopathological specimens, serving to preserve tissue structure and prevent autolysis. The most commonly used fixative is 10% Neutral Buffer Formalin (NBF), although it is known to be toxic and carcinogenic. This study aims to observe the microscopic appearance of chicken liver tissue fixed using 10% NBF and honey (Dimocarpus Longan) as a natural alternative fixative at concentrations of 5%, 15%, and 100%. The study was conducted experimentally in the Anatomical Pathology Laboratory of RSUD Dr. Soeratno Gemolong, using broiler chicken liver tissue fixed for 24 hours. Fixation was performed using 10% Neutral Buffer Formalin (NBF) and Longan honey (Dimocarpus longan) at concentrations of 5%, 15%, and 100%. Following histotechnical processing and Hematoxylin-Eosin (HE) staining, microscopic examination was carried out. The results showed that liver tissue fixed with 10% NBF exhibited well- preserved microscopic structure with intact cells. In contrast, tissues fixed with 5% and 15% honey were destroyed during fixation. Honey at 100% concentration yielded suboptimal fixation, showing necrotic tissue, pyknotic nuclei, and incomplete preservation. In conclusion, honey has not been proven effective as a substitute for 10% NBF in preserving the microscopic structure of chicken liver tissue. The fixation results were inadequate, particularly at lower concentrations. Further research is recommended to explore other natural sweeteners or substances as potential fixative alternatives.
