Abstract
Hiperglikemia ialah kondisi kadar gula darah tinggi, dalam
jangka panjang dapat menyebabkan kerusakan fungsional berbagai
organ. Pucuk daun mangga arum manis memiliki aktivitas
antihiperglikemia sebab mengandung flavonoid. Tujuan penelitian
untuk mengetahui aktivitas antihiperglikemia ekstrak tumbuhan
tersebut terhadap penurunan kadar glukosa darah pada mencit putih
jantan yang diinduksi aloksan dan mengetahui dosis efektif
tumbuhan tersebut.
Ekstraksi serbuk tumbuhan ini dilakukan dengan metode
maserasi menggunakan pelarut etanol 70%. Penelitian menggunakan
25 ekor mencit terbagi dalam 6 kelompok perlakuan dengan aloksan
sebagai penginduksi. Kelompok perlakuan terbagi menjadi: (ekstrak
dengan dosis 52,5mg/kgBB, 105mg/kgBB, 210 mg/kgBB), kontrol
negatif (Na CMC), dan kelompok normal dilakukan uji selama 14
hari. Pengukuran kadar glukosa menggunakan alat glukometer
sebelum induksi aloksan (T0), setelah pemberian aloksan (T1), dan
sesudah diberi perlakuan (T2 dan T3). Mencit selanjutnya
dikorbankan dan diambil pankreasnya untuk diuji histopatologi. Data
yang dihasilkan diuji menggunakan wilcoxon, anova, tukey HSD, dan
dunnett T3.
Hasil yang didapatkan pada penelitian bahwa ekstrak pucuk
daun mangga arum manis dengan berbagai variasi dosis dapat
menurunkan kadar gula darah. Dosis ekstrak yang mampu
memberikan penurunan kadar glukosa darah yang paling efektif yaitu
dosis 52,5 mg/kgBB.
Hyperglycemia is a condition of high blood sugar levels, in
the long term it can cause functional damage to various organs. Sweet
mango leaf tips have antihyperglycemic activity because they contain
flavonoids. The purpose of this study was to determine the
antihyperglycemic activity of the plant extract on reducing blood
glucose levels in male white mice induced by alloxan and to
determine the effective dose of the plant.
Extraction of this plant powder was carried out using the
maceration method using 70% ethanol solvent. The study used 25
mice divided into 6 treatment groups with alloxan as an inducer. The
treatment groups were divided into: (extract with a dose of 52.5 mg /
kgBW, 105 mg / kgBW, 210 mg / kgBW), negative control (Na
CMC), and the normal group was tested for 14 days. Measurement
of glucose levels using a glucometer before alloxan induction (T0),
after alloxan administration (T1), and after treatment (T2 and T3).
The mice were then sacrificed and their pancreas was taken for
histopathological testing.
The resulting data were tested using Wilcoxon, ANOVA,
Tukey HSD, and Dunnett T3. The results obtained in the study
showed that the extract of sweet mango leaf shoots with various doses
can reduce blood sugar levels. The dose of extract that can provide
the most effective decrease in blood glucose levels is a dose of 52.5
mg/kgBW.
