UJI EFEKTIVITAS FRAKSI EKSTRAK ETANOL DAUN LAMTORO (Leucaena leucocephala L.) SEBAGAI ANTIDIABETES PADA MENCIT PUTIH JANTAN (Mus musculus)

Diabetes mellitus sebagai salah satu penyakit degeneratif merupakan kondisi jangka panjang yang ditandai dengan peningkatan kadar gula darah dan gangguan metabolisme. Daun lamtoro merupakan salah satu tanaman yang sering digunakan sebagai obat tradisional. Tujuan penelitian ini adalah mengetahui efektivitas fraksi ekstrak etanol dari daun lamtoro dan dosis efektif dari fraksi ekstrak etanol daun lamtoro sebagai antidiabetes, serta gambaran histopatologi pankreas pada mencit jantan yang diinduksi aloksan. Penelitian ini menggunakan 30 ekor mencit jantan yang dibagi menjadi 6 kelompok, setiap kelompok terdiri dari 5 ekor mencit jantan. Kontrol positif glibenklamid, kontrol negatif Na CMC 0,5%, ekstrak etanol daun lamtoro 150 mg/kg BB, fraksi n-heksana daun lamtoro 60 mg/kg BB, fraksi etil asetat daun lamtoro 30 mg/kg BB, dan fraksi air daun lamtoro 75 mg/kg BB. Semua kelompok diberi perlakuan selama 13 hari. Hari ke-0, hari ke-3, hari ke-7, dan hari ke-13 menetapkan kadar gula darah, pada hari ke-14 mencit dibedah kemudian diambil organ pankreas yang digunakan sebagai preparat histopatologi. Kadar gula darah pada mencit diukur menggunakan glukometer melalui intravena. Data yang diperoleh selanjutnya dianalisis dengan menggunakan aplikasi SPPS dengan metode One Way Anova kemudian dilanjutkan uji Post Hoc. Hasil penelitian menunjukkan bahwa fraksi ekstrak etanol daun lamtoro dapat menurunkan kadar gula darah pada mencit. Fraksi ekstrak etanol daun lamtoro efektif dapat menurunkan kadar gula darah pada dosis 75 mg/kg BB fraksi air, 30 mg/kg BB fraksi etil asetat, dan 60 mg/kg BB fraksi n-heksana. Pemberian dosis fraksi ekstrak etanol daun lamtoro pada gambaran histopatologi pankreas dapat menurunkan kerusakan sel nekrotik mencit. Diabetes mellitus as one of the degenerative diseases is a long�term condition characterized by increased blood sugar levels and metabolic disorders. Lamtoro leaves are one of the plants that are often used as traditional medicine. The purpose of this study was to determine the effectiveness of the ethanol extract fraction of lamtoro leaves and the effective dose of the ethanol extract fraction of lamtoro leaves as an antidiabetic, as well as the histopathological picture of the pancreas in male mice induced by alloxan. This study used 30 male mice divided into 6 groups, each group consisting of 5 male mice. Positive control glibenclamide, negative control Na CMC 0.5%, ethanol extract of lamtoro leaves 150 mg/kg BW, n-hexane fraction of lamtoro leaves 60 mg/kg BW, ethyl acetate fraction of lamtoro leaves 30 mg/kg BW, and water fraction of lamtoro leaves 75 mg/kg BW. All groups were treated for 13 days. Day 0, day 3, day 7, and day 13 determine blood sugar levels, on day 14 mice were dissected and pancreas organs were taken which were used as histopathology preparations. Blood sugar levels in mice were measured using a glucometer via intravenous. The data obtained were then analyzed using the SPPS application with the One Way Anova method followed by the Post Hoc test. The results showed that the fraction of lamtoro leaf ethanol extract can reduce blood sugar levels in mice. Fractions of lamtoro leaf ethanol extract can effectively reduce blood sugar levels at doses of 75 mg/kg BW of water fraction, 30 mg/kg BW of ethyl acetate fraction, and 60 mg/kg BW of n-hexane fraction. Dosing the fraction of lamtoro leaf ethanol extract on the histopathology picture of the pancreas can reduce necrotic cell damage to mice.
