Abstract
Tanaman jeruk purut merupakan tanaman yang sering
digunakan oleh masyarakat sebagai ramuan obat tradisional.
Banyaknya perkembangan penggunaan obat tradisional di kalangan
masyarakat mendorong penelitian terkait prediksi profil
farmakokinetika dan toksisitas yang masih belum dilakukan..
Penelitian ini menggunakan webserver ADMETlab.
Pengumpulan data senyawa menggunakan webserver KNApSAcK.
Pengumpulan kode smile dilakukan dengan mengakses webserver
PubChem. Seleksi kemiripan obat dilakukan pada webserver
SwissADME . Pengumpulan data farmakokinetika ADME dan
toksisitas menggunakan ADMETlab 3.0. Analisis dilakukan manual
dengan membandingkan data yang diperoleh dengan keputusan empiris
tiap parameter yang telah ada.
Hasil penelitian menunjukan hanya 36% senyawa yang lolos
pengujian druglikeness. Analisis ADME menunjukan karakteristik
berbeda berdasarkan golongan senyawa, hal ini dipengaruhi oleh sifat
fisikokimia dan gugus fungsi spesifik dari kandungan senyawa
golongan. Analisis toksisitas senyawa jeruk purut menunjukan
berpotensi toksik terutama karsinogenitasnya dan pada TOX21
pathway beberapa senyawa golongan terpenoid cenderung bersifat
tidak aktif dibandingkan golongan senyawa lainya. Prediksi kandidat
obat terbaik yang memiliki profil farmakokinetika dan toksisitas adalah
senyawa citronelyll formate.
Kaffir lime plant is a plant that is often used by the community
as a traditional medicinal herb. The number of developments in the use
of traditional medicine among the community encourages research
related to the prediction of pharmacokinetic and toxicity profiles that
have not yet been carried out.
This research uses ADMETlab webserver. Compound data
collection using the KNApSAcK webserver. Smile code collection was
done by accessing the PubChem webserver. Drug similarity selection
was performed on the SwissADME webserver. ADME
pharmacokinetics and toxicity data were collected using ADMETlab
3.0. The analysis was done manually by comparing the data obtained
with the empirical decision for each parameter that already exists.
The results showed that only 36% of the compounds passed
druglikeness testing. ADME analysis shows different characteristics
based on the compound group, this is influenced by the
physicochemical properties and specific functional groups of the
compound group content. Toxicity analysis of kaffir lime compounds
shows toxic potential, especially carcinogenicity and in the TOX21
pathway some terpenoid group compounds tend to be inactive
compared to other compound groups. The best predicted drug
candidate that has the best pharmacokinetics and toxicity profile is the
compound citronelyll formate.
