Abstract
Bakteri endofit merupakan mikroba endofit yang hidup dalam
jaringan tanaman, yang dapat menghasilkan senyawa bioaktif sebagai
antibakteri. Bunga kembang sepatu (Hibiscus rosa sinensis L.) memiliki
senyawa metabolit sekunder yang berkhasiat sebagai antibakteri. Tujuan
penelitian ini adalah mengidentifikasi genus bakteri endofit dan
mengetahui aktivitas antibakteri terhadap bakteri Staphylococcus
aureus.
Bunga kembang sepatu dilakukan sterilisasi permukaan dan
diisolasi untuk mendapatkan isolat murni bakteri endofit. Identifikasi
genus bakteri berdasarkan uji morfologi, pewarnaan Gram, dan uji
biokimia. Bakteri endofit difermentasi dengan perbedaan waktu
fermentasi 24, 48, dan 72 jam, hasil fermentasi dilakukan sentrifugasi
untuk mendapatkan supernatan yang digunakan dalam pengujian
aktivitas antibakteri menggunakan metode difusi cakram.
Hasil identifikasi genus diperoleh 6 genus bakteri endofit
diantaranya genus Escherichia, Aeromicrobium, Pseudomonas,
Paenibacillus, Bacillus, dan Klebsiella. Hasil uji aktivitas antibakteri
menunjukkan 5 isolat bakteri dengan waktu fermentasi 48 dan 72 jam
menunjukkan diameter zona hambat terhadap bakteri S. aureus. Isolat
bakteri endofit bunga kembang sepatu yang memiliki diameter zona
hambat paling tinggi pada waktu fermentasi 72 jam dengan nilai 10,22
mm yaitu genus Klebsiella. Waktu fermentasi 72 jam menghasilkan
diameter daya hambat yang tinggi yang dipengaruhi oleh metabolit
sekunder yang diproduksi isolat bakteri endofit.
Endophytic bacteria are endophytic microbes that live in plant
tissues, which can produce bioactive compounds as antibacterials.
Hibiscus flower (Hibiscus rosa sinensis L.) has secondary metabolite that
have antibacterial properties. The purpose of this study was to identify
the genus of endophytic bacteria and determine the antibacterial activity
against Staphylococcus aureus bacteria.
Hibiscus flowers were sterilized and isolated to obtain pure
isolates of endophytic bacteria. Identification of the bacterial genus was
based on morphological tests, Gram staining, and biochemical tests.
Endophytic bacteria were fermented with different fermentation times of
24, 48, and 72 hours, the fermentation results were centrifuged to obtain
supernatants used in testing antibacterial activity using the disc diffusion
method.
The results of genus identification obtained 6 genus of
endophytic bacteria including the genus Escherichia, Aeromicrobium,
Pseudomonas, Paenibacillus, Bacillus, and Klebsiella. The results of the
antibacterial activity test showed that 5 bacterial isolates with
fermentation times of 48 and 72 hours showed the diameter of the
inhibition zone against S. aureus bacteria. The hibiscus flower
endophytic bacterial isolate that has the highest inhibition zone diameter
at 72 hoursfermentation time with a value of 10.22 mm is IBKS 6 isolate.
72 hours fermentation time produces a high inhibition diameter which is
influenced by secondary metabolites produced by endophytic bacterial
isolates.
