Abstract
Minuman dingin banyak dikonsumsi masyarakat Indonesia. Es
kristal dan es batu menjadi pilihan untuk mendinginkan minuman. Es
kristal dan es batu berbahan baku air yang menjadi sumber kontaminasi
bakteri koliform. Koliform memiliki potensi untuk menyebabkan
penyakit seperti diare, tifus, dan disentri basiler. Tujuan penelitian ini
adalah untuk mengetahui perbandingan bakteri koliform es kristal dan es
batu yang beredar di salah satu kantin kampus swasta kota Surakarta.
Sampel es kristal dan es batu dilakukan uji Angka Paling Mungkin
(APM). Penyiapan uji praduga menggunakan media Lactose Broth (LB)
dengan ragam 333. Hasil uji Praduga yang positif diuji ke media Brilliant
Green Lactose Bile Broth (BGLB) dengan diambil 1-2 ose secara aseptis.
Uji Pelengkap menggunakan Mac Conkey Agar dengan teknik gores dari
hasil uji penegasan serta identifikasi bakteri dengan pewarnaan Gram
dan uji biokimia (SIM, KIA, LIA, Sitrat).
Berdasarkan hasil penelitian es kristal dengan ragam 333 terdapat
kekeruhan dan gas pada tabung durham kecuali pada satu tabung durham
di pemipetan sampel 1 mL dan memperoleh nilai APM koliform yaitu
29.000/100 mL sedangkan pada es batu terdapat kekeruhan dan gas di
semua tabung durham maka memperoleh nilai APM koliform
>110.000/100 mL. APM koliform es kristal lebih kecil daripada APM
koliform es batu. Es kristal dan es batu yang beredar di salah satu kantin
kampus swasta kota Surakarta tidak memenuhi syarat mutu SNI
3839:2019. Cemaran bakteri dalam es kristal serta es batu yaitu
Pseudomonas dan Klebsiella sebagai bakteri koliformnya
Cold drinks are popular among Indonesians, who often use crystal
ice and ice cubes to cool their beverages. These ice varieties are produced
using water, which can potentially be contaminated with coliform
bacteria. Coliform bacteria have the potential to cause diseases such as
diarrhea, typhoid fever, and bacillary dysentery. The purpose of this
research is to determine the comparison of coliform bacteria in crystal
ice and ice cubes circulating in one of the private university canteens in
Surakarta city.
Samples of crystal ice and ice cubes underwent Most Probable
Number (MPN). Presumptive testing was using Lactose Broth (LB) with
strain 333. Positive presumptive results were confirmed using Brilliant
Green Lactose Bile Broth (BGLB) with 1-2 ml ose aseptically. Further
test were performed using Mac Conkey Agar with streaking techniques
based on confirmatory test results. Bacteria identification with gram
staining and biochemical tests (SIM, KIA, LIA, Citrate).
Based on the research results, crystal ice with variety 333 showed
turbidity and gas in Durham tubes except for one Durham tube in the
pipetting of 1 mL sample and obtained coliform MPN values of
29,000/100 mL, while ice cubes showed turbidity and gas in all Durham
tubes, resulting in coliform MPN values >110,000/100 mL. The coliform
MPN value in crystal ice is lower than in ice cubes. The crystal ice and
ice cubes sold at a private university canteen in Surakarta city do not
comply with the quality standards set by SNI 3839:2019. Bacterial
contamination in crystal ice and ice cubes includes Pseudomonas and
Klebsiella as coliform bacteria.
