Abstract

Nifedipin merupakan salah satu obat untuk hipertensi yang termasuk golongan calcium channel blockers (CCB) jenis dihidropiridin dengan bioavailabilitas rendah. Self Nano-Emulsifying Drug Delivery System (SNEDDS) adalah formula berbasis lipid dengan campuran isotropik dari minyak alami maupun sintetik, surfaktan, dan kosurfaktan dengan pengadukan ringan yang meningkatkan bioavailabilitas oral obat yang sukar larut dalam air. Tujuan penelitian adalah mengetahui pengaruh minyak, surfaktan, dan kosurfaktan dalam terbentuknya SNEDDS nifedipine, karakterisasi SNEDDS nifedipine dengan variasi konsentrasi asam oleat, dan transcutol, perbandingan optimum minyak : kosurfaktan formulasi SNEDDS nifedipine menggunakan metode SLD. Penelitian pembuatan SNEDDS nifedipine menggunakan kombinasi asam oleat (fase minyak) : transcutol (kosurfaktan) dengan jumlah tween 80 sebagai surfaktan yang sama. Kombinasi asam oleat dan transcutol dimasukkan dalam design expert dihasilkan 8 run dengan komposisi R1 (1:1), R2 (1:1), R3 (1=0,75:1,25), R4 (1,25:0,75), R5 (0,5:1,5), R6 (1,5:0,5), R7 (1,5:0,5), R8 (0,5:1,5), dan ditambahkan nifedipine sebagai zat aktif. Hasil data 8 run yang diperoleh dari uji karakteristik dan kemudian dimasukkan dalam software Design Expert dengan program SLD (Simplex Lattice Design), sehingga didapatkan hasil yang optimum untuk SNEDDS Nifedipine. SLD memiliki rumus terbaik untuk campuran bahan dimana proporsi jumlah total bahan yang berbeda harus 1 (100%) Hasil karakterisasi optimum secara prediksi menggunakan SLD dengan minyak minyak 10,162 % dan kosurfaktan 29,838 % akan menghasilkan waktu emulsifikasi sebesar 26,379 detik, drug loading 102,162 ppm, dan % transmitan sebesar 64,190 %. Hasil penelitian menunjukkan setelah dilakukan verivikasi karakterisasi didapatkan hasil waktu emulsifikasi sebesar 26,00 detik, drug loading 101,35 ppm, dan % transmitan sebesar 64,11 %. Nifedipine is one of the drugs for hypertension which belongs to the class of calcium channel blockers (CCB) type of dihydropyridine with low bioavailability. The Self Nano-Emulsifying Drug Delivery System (SNEDDS) is a lipid-based formula with an isotropic blend of natural and synthetic oils, surfactants, and cosurfactants with mild stirring that improves the oral bioavailability of water-insoluble drugs. The purpose of the study was to determine the effect of oil, surfactant, and cosurfactant in the formation of nifedipine SNEDDS, characterization of nifedipine SNEDDS with variations in oleic acid concentration, and transcutol, optimum ratio of oil: cosurfactant formulation of nifedipine SNEDDS using the SLD method. This study made 8 runs SNEDDS using tween 80 as a surfactant of 3 grams with a ratio of 8 formulas of oleic acid (oil phase): transcutol (cosurfactant); R1 (1:1), R2 (1:1), R3 (1=0.75:1.25), R4 (1.25:0.75), R5 (0.5:1.5), R6 (1.5:0.5), R7 (1.5:0.5), R8 (0.5:1.5), and nifedipine is added as an active substance. The results of 8 runs of data obtained from characteristic tests and then entered in the Design Expert software with the SLD (Simplex Lattice Design) program, so that optimal results are obtained for SNEDDS Nifedipine. SLD has the best formula for material mixtures where the proportion of the total amount of different ingredients should be 1 (100%) The results of the optimal characterization predictably using SLD with oil oil of 10.162% and cosurfactant of 29.838% will result in an emulsification time of 26.379 seconds, a drug loading of 102.162 ppm, and a transmittance % of 64.190%. The results of the study showed that after the characterization verification was carried out, the results of emulsification time of 26.00 seconds, drug loading of 101.35 ppm, and % transmittance of 64.11 % were obtained.
