Abstract
Tablet lepas lambat kaptopril sediaan yang memiliki pelepasan obat yang relatif lama. Tablet lepas lambat dirancang untuk melepaskan obat secara bertahap agar memperpanjang kerja obat. Penelitian ini bertujuan untuk melihat pengaruh tambahan matriks HPMC K4M dan Na. CMC terhadap mutu fisik dan disolusi sediaan lepas lambat kaptopril, serta dengan konsentrasi HPMC K4M dan Na. CMC untuk mengetahui sediaan lepas lambat kaptopril yang optimum.
Pembuatan sediaan menggunakan metode granulasi basah dan menggunakan 8 formula dari Simplex Lattice Design (SLD) dengan perbandingan HPMC K4M dan Na. CM. Uji mutu fisik dengan parameter kritis kekerasan, kerapuhan dan DE360 dianalisis dengan Software Simplex Lattice Design (SLD). Metode ini dapat digunakan sebagai formula sediaan dengan berbagai variasi jumlah komposisi bahan, sehingga mendapatkan formula optimum yang diharapkan.
HPMC K4M dan Na.CMC perpengaruh paling dominan terhadap penurunan kecepatan disolusi dan interaksi keduanya jika dikombinasikan akan berpengaruh terhadap kekerasan tablet. Berdasarkan Software Simplex Lattice Design (SLD) diperoleh daerah optimum dengan komposisi HPMC K4M 4,000 mg dan Na.CMC 12,000 mg menghasilkan nilai kekersan, kerapuhan dan DE360 yang baik.
Captopril extended-release tablets are a preparation that has a relatively long release of the drug. Sustained-release tablets are designed to release the drug gradually to prolong the action of the drug. This research aims to see the effect of additional HPMC K4M and Na matrices. CMC on physical quality and dissolution of captopril extended release preparations, as well as HPMC K4M and Na concentrations. CMC to determine the optimum slow release preparation of captopril.
The preparation uses the wet granulation method and uses 8 formulas from Simplex Lattice Design (SLD) with a ratio of HPMC K4M and Na. C.M. Physical quality tests with critical parameters of hardness, brittleness and DE360 were analyzed using Simplex Lattice Design (SLD) Software. This method can be used as a preparation formula with various variations in the amount of ingredient composition, so as to obtain the desired optimum formula.
HPMC K4M and Na.CMC have the most dominant influence on reducing dissolution speed and the interaction of the two when combined will affect tablet hardness. Based on the Simplex Lattice Design (SLD) software, the optimum area with a composition of 4,000 mg HPMC K4M and 12,000 mg Na.CMC produces good hardness, brittleness and DE360 values.
