Abstract
Hiperglikemia adalah suatu kondisi dimana kadar glukosa darah
meningkat melampaui batas normal yang merupakan ciri dari beberapa
penyakit, terutama diabetes melitus. Salah satu tumbuhan yang dapat
digunakan untuk penurunan hiperglikemik yaitu daun pare (Momordica
charantia L.). Peneliti bertujuan untuk mengetahui aktivitas
antihiperglikemik, dosis efektif serta untuk mengetahui kemampuan
ekstrak etanol daun pare dalam memperbaiki histopatologi pankreas
pada mencit yang diinduksi STZ-NA.
Penelitian ini ekstrak daun pare diperoleh menggunakan metode
maserasi dengan pelarut etanol 96%. Hewan uji yang digunakan 24
mencit jantan dibagi 6 kelompok perlakuan yaitu kelompok I (normal),
kelompok II (negatif), kelompok III (positif), kelompok IV (ekstrak
dosis 70 mg/kgBB), kelompok V (ekstrak dosis 140 mg/kgBB) dan
kelompok VI (ekstrak dosis 280 mg/kgBB). Hewan uji dibuat
hiperglikemik dengan diinduksi STZ-NA dan diukur kadar glukosa
darah mencit pada hari ke-0, 3 dan 10 dan 17 dengan glukometer dan
dilakukan uji histopatologi organ pankreas mencit. Data penurunan
kadar glukosa darah dan histopatologi pankreas dianalisis bahwa data
terdistribusi normal dan homogen, dilanjutkan dengan uji One-Way
ANOVA kemudian dilanjutkan dengan uji Post Hoc Tukey HSD untuk
melihat perbedaan antar kelompok.
Hasil penelitian menunjukan bahwa pemberian ekstrak etanol daun
pare dapat memberikan aktivitas antihiperglikemik, dosis efektif ekstrak
etanol daun pare yang dapat memberikan aktivitas antihiperglikemik
adalah dosis 140 mg/kgBB mencit dan ekstrak etanol daun pare dapat
memperbaiki histopatologi pankreas pada mencit yang diinduksi STZNA.
Hyperglycemia is a condition where blood glucose levels increase
beyond normal limits which is a characteristic of several diseases,
especially diabetes mellitus. One plant that can be used to reduce
hyperglycemia is bitter melon leaves (Momordica charantia L.).
Researchers aim to determine antihyperglycemic activity, effective
dosage and to determine the ability of bitter melon leaf ethanol extract to
improve pancreatic histopathology in STZ-NA induced mice.
In this research, bitter melon leaf extract was obtained using the
maceration method with 96% ethanol solvent. The test animals used
were 24 male mice divided into 6 treatment groups, namely group I
(normal), group II (negative), group III (positive), group IV (extract dose
70 mg/kgBB), group V (extract dose 140 mg/kgBB ) and group VI
(extract dose 280 mg/kgBB). The test animals were made hyperglycemic
by being induced by STZ-NA and the blood glucose levels of the mice
were measured on days 0, 3, 10 and 17 using a glucometer and
histopathological tests were carried out on the mice's pancreas. Data on
decreased blood glucose levels and pancreatic histopathology were
analyzed to show that the data were normally distributed and
homogeneous, followed by the One-Way ANOVA test then followed by
the Post Hoc Tukey HSD test to see differences between groups.
The results of the research show that administration of bitter melon
leaf ethanol extract can provide antihyperglycemic activity. The
effective dose of bitter melon leaf ethanol extract which can provide
antihyperglycemic activity is a dose of 140 mg/kgBW of mice and bitter
melon leaf ethanol extract can improve pancreatic histopathology in
mice induced by STZ-NA.
