Abstract
Tonikum merupakan sediaan atau obat penambah daya tahan
tubuh. Herba sambiloto mengandung senyawa saponin, flavonoid,
alkaloid, tanin dan senyawa diterpen yang komponen utamanya adalah
andrographolide. Penelitian ini bertujuan untuk mengetahui efek
tonikum, fraksi teraktif dari fraksi air, etil asetat dan n-heksana herba
sambiloto serta golongan senyawa dalam fraksi teraktif.
Serbuk herba sambiloto dimaserasi menggunakan etanol 70%,
dan difraksinasi dengan menggunakan air, etil asetat, dan n-heksana.
Uji aktivitas tonikum menggunakan metode gelantung dengan hewan
uji mencit 24 ekor. Kontrol negatif Na-CMC 0,5 (I), kontrol positif
kafein 100 mg/KgBB (II), ekstrak herba sambiloto dosis 74,554
mg/KgBB (III), fraksi n-heksana dengan dosis 3,18 mg/KgBB (IV),
fraksi etil asetat dengan dosis 3,67 mg/KgBB (V), fraksi air dengan
dosis 44,08 mg/KgBB (VI). Hasil data yang diperoleh pada uji efek
tonikum dari selisih waktu sebelum pemberian (T0) dan sesudah
pemberian (T1) dianalisis statistik dengan menggunakan One-way
ANOVA.
Fraksi n-heksana, fraksi etil asetat, dan fraksi air mempunyai
aktivitas tonikum. Fraksi etil asetat dengan dosis 3,67 mg/KgBB
merupakan fraksi teraktif dan memiliki efek tonikum sebanding dengan
kafein 100 mg/KgBB. Senyawa flavonoid dan alkaloid yang
terkandung dalm fraksi teraktif diduga berperan atas efek tonikum.
Tonics are preparations or drugs that increase endurance.
Sambiloto herb contains saponin, flavonoid, tanin and diterpene
compounds whose main component is andrographolide. This research
aims to determine the tonic effect, the most active fraction from the
water fraction, ethyl acetate and n-hexane of the bitter herbs as well as
the group of compound in the active fraction.
Sambiloto herb powder was macerated using 70% ethanol, and
fractionated using water, ethyl acetate, and n-hexane. The tonic activity
test used the hanging method with 24 mice as test animal. Negative
control Na-CMC 0,5 % (I), positive control caffeine 100 mg/KgBB (II),
sambiloto herb extract with a dose of 74,554 mg/KgBB (III), n-hexane
fraction with a dose of 3,18 mg/KgBB (IV), ethyl acetate fraction with
a dose of 3,67 mg/KgBB (V), water fraction with a dose of 44,08
mg/KgBB (VI). The results of the data obtained in the tonic effect test
from the time difference before administration (T0) and after
administration (T1) were analyzed statistically using One Way ANOVA.
The n-hexane fraction, ethyl acetate fraction, and water fraction
have tonic activity. The ethyl acetate fraction with a dose of 3,67
mg/KgBB is the most active fraction and has a tonic effect comparable
to caffeine 100 mg/KgBB. The flavonoid and alkaloid compound
contained in the active fraction are though to play a role in the tonic
effect.
