Abstract

Histologi adalah ilmu yang mempelajari tentang morfologi dan sifat jaringan atau sel yang normal. Histoteknik adalah metode untuk membuat preparat histologi dari spesimen tertentu melalui serangkaian proses hingga menjadi preparat yang siap untuk diamati atau dianalisis menggunakan mikroskop. Hematoxylin dan Eosin yang bertujuan untuk melihat bentuk sel seperti inti, sitoplasma dan bagian lainnya pada jaringan dan sangat diperlukan dalam diagnosa medis dan penelitian. Tujuan dari penelitian ini untuk mengetahui perbedaan hasil pewarnaan gambaran preparat histologi inti sel dan sitoplasma sel hepar mencit dengan perbedaan pewarnaan Hematoxylin Eosin. Penelitian di lakukan di laboratorium patologi anatomi RSUD dr. Soeratno Gemolong, pada bulan mei 2023. Sampel pada penelitian ini adalah 6 organ hepar atau blok Histologi, masing-masing di buat blok jaringan dan dibagi menjadi 2. 3 sediaan untuk Harris dan 3 sediaan untuk Mayer setelah itu di lakukan uji coba setelah itu diamati dibawah mikroskop. Kesimpulan dari penelitian ini berdasarkan hasil pengamatan pada penelitian dan pembahasan dapat disimpulkan bahwa hasil mikroskopis jaringan histologi yang diwarnai dengan pewarnaan Hematoxylin Eosin yang menggunakan pewarnaan Hematoxylin Mayer dan Hematoxylin Harris. Hematoxylin Mayer inti terwarnai berwarna ungu dan bagian inti terlihat sangat jelas. Hematoxylin Harris inti terwarnai berwarna biru keungguan dan bagian inti terlihat kurang jelas. Histology is the science that studies the morphology and properties of normal tissue or cells. Histotechnics is a method for making histological preparations from certain specimens through a series of processes until they become preparations that are ready to be observed or analyzed using a microscope. Hematoxylin and Eosin which aims to see the shape of cells such as the nucleus, cytoplasm and other parts of tissue and are very necessary in medical diagnosis and research. The aim of this study was to determine the differences in staining results of histological preparations of cell nuclei and cytoplasm of mouse liver cells with differences in Hematoxylin Eosin staining. The research was carried out in the anatomical pathology laboratory at RSUD dr. Soeratno Gemolong, in May 2023. The samples in this study were 6 liver organs or Histology blocks, each tissue block was made and divided into 2. 3 preparations for Harris and 3 preparations for Mayer after that a trial was carried out and then observed under the microscope. The conclusion of this study is based on the results of observations in the research and discussion that it can be concluded that the microscopic results of histological tissue stained with Hematoxylin Eosin staining using Hematoxylin Mayer and Hematoxylin Harris staining. Mayer's hematoxylin nuclei are stained purple and the nuclei are clearly visible. Hematoxylin Harris nuclei are stained purplish blue and the nuclei are less clearly visible.
