UJI AKTIVITAS EKSTRAK DAN FRAKSI BIJI KOPI ROBUSTA (Coffea canephora) SEBAGAI ANTIHIPERGLIKEMIK DAN REGENERASI SEL BETA PANKREAS

Diabetes Melitus (DM) merupakan penyakit yang terjadi karenakan gangguan metabolik. Hormon insulin mengalami gangguan disekresi insulin pada sel beta langerhans atau berkurangnya sensitivitas insulin terhadap reseptor. sehingga mengakibatkan terjadinya hiperglikemik. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antihiperglikemik dari ekstrak dan fraksi biji kopi robusta serta dosis optimal fraksi yang digunakan sebagai antihiperglikemik dan histolopatologi pankreas. Penelitian ini menggunakan hewan uji mencit dan tikus yang dikondisikan dengan induksi STZ-NA. Mengunakan 30 ekor mencit kemudian di kelompokan menjadi 6 kelompok. Ekstrak etanol biji kopi robusta 400mg/kg BB mencit, fraksi n-heksan 42 mg/kg BB , fraksi etil asetat 37,4 mg/kg BB mencit, fraksi air 216 mg/kg BB mencit, kontrol positif dan kontrol negatif. Dilakukan pengujian antihiperglikemik, selanjutnya hewan uji tikus yang digunakan 35 ekor yang kemudian dibagi dalam 7 kelompok. Dosis streptozotosin 40 mg/kg BB nikotinamid 110 mg/kg BB, ekstrak biji kopi robusta 280 mg/kg BB, fraksi etil asetat 13,3 mg/kg BB, fraksi etil asetat 26,6 mg/kg BB, fraksi etil asetat 53,2 mg/kg BB, kontrol positif, kontrol negatif dan kontrol normal. Dilakukan uji aktivitas antihiperglikemik dan histopatologi pankreas. Hasil penelitian menunjukan bahwa ekstrak etanol biji kopi robusta memiliki aktivitas antihiperglikemik. Fraksi etil asetat biji kopi robusta memiliki Dosis 26,6 mg/kg BB merupakan dosis optimal sebagai antihiperglikemik dan meningkatkan regenerasi pulau Langerhans. This research used test animals of mice and rats conditioned with STZ-NA induction. Using 30 mice then grouped into 6 groups. Robusta coffee bean ethanol extract 400 mg/kg BW of mice, n-hexane fraction 42 mg/kg BW, ethyl acetate fraction 37,4 mg/kg BW of mice, water fraction 216 mg/kg BW of mice, positive control and negative control. Antihyperglycemic testing was carried out, then 35 mice were used as test animals which were then divided into 7 groups. Streptozotocin dose 40 mg/kg BW nicotinamide 110 mg/kg BW, robusta coffee bean extract 280 mg/kg BW, ethyl acetate fraction 13,3 mg/kg BW, ethyl acetate fraction 26,6 mg/kg BW, ethyl acetate fraction 53,2 mg/kg BW, positive control, negative control and normal control. Antihyperglycemic activity and pancreati histopathology tests were carried out. The research results show that the ethanol extract of robusta coffee beans has antihyperglycemic activity. The ethyl acetate fraction of robusta coffee beans has a dose of 26,6 mg/kg BW, which is the optimal dose as an antihyperglycemic and increases the regeneration of the islets of Langerhans.
