UJI TOKSISITAS SUBKRONIS FRAKSI DENGAN TITIK DIDIH TERTINGGI DARI MINYAK PALA TERHADAP OTAK DAN PANKREAS TIKUS PUTIH (Rattus novergicus) GALUR WISTAR

Miristisin memiliki berbagai aktivitas diantaranya antioksidan, antibakteri, antiinflamasi, dan antidepresan. Informasi mengenai pengaruh efek toksik miristisin terhadap organ tikus putih (Rattus novergicus) belum ditemukan. Tujuan dari penelitian ini untuk mengetahui efek toksik isolat miristisin dari tanaman pala (Myristica fragrans Houtt) dan mengetahui pengaruh efek toksik dari isolat miristisin terhadap organ otak dan pankreas tikus putih (Rattus novergicus). Sampel yang digunakan adalah miristisin hasil isolasi dari penelitian sebelumnya. Metode yang digunakan adalah subkronis oral dengan pemberian dosis berulang selama 28 hari. Kelompok uji terbagi menjadi 7 kelompok, yaitu kelompok kontrol normal, dosis 2,1;27,3;3,549 mg/kg BB, kontrol negatif, dan kelompok satelit yang terdiri dari kontrol satelit dan dosis 354,9 mg/kg BB. Parameter uji yang diamati antara lain gejala toksik; makropatologi organ; bobot organ relatif; pemeriksaan histopatologi. Analisis data menggunakan uji Shapiro-Wilk dan uji Homogenity of Variances dilanjutkan dengan uji one way ANOVA dan kelompok satelit menggunakan uji Independent Sample Test. Data yang tidak terdistribusi normal dianalisis menggunakan Kruskal Wallis dan Mann Withney. Hasil penelitian menyatakan dosis toksik miristisin (LD₅₀) yaitu 9,7813 mg/kg BB tikus menyebabkan kematian dan berpengaruh terhadap otak dan pankreas pada dosis 27,3 mg/kg BB dan 354,9 mg/kg BB yang ditandai dengan adanya edema dan kongesti ringan hingga sedang pada otak, dan edema ringan pada pankreas. Information regarding the toxic effects of myristicin on the organs of white rats (Rattus novergicus) has not been found. The aim of this research was to determine the toxic effect of myristicin isolate from the nutmeg plant (Myristica fragrans Houtt) and to determine the effect of the toxic effect of myristicin isolate on the brain and pancreas of white rats (Rattus novergicus). The sample used was myristicin isolated from previous research. The method used is subchronic oral administration with repeated doses for 28 days. The test group was divided into 7 groups, namely the normal control group, dose 2.1; 27.3; 3,549 mg/kg BW, negative control, and the satellite group consisting of satellite control and dose 354.9 mg/kg BW. The test parameters observed included toxic symptoms; organ macropathology; relative organ weights; histopathological examination. Data analysis used the Shapiro-Wilk test and the Homogeneity of Variances test followed by the one way ANOVA test and the satellite group used the Independent Sample Test. Data that were not normally distributed were analyzed using Kruskal Wallis and Mann Withney. The results of the study stated that the toxic dose of myristicin (LD₅₀), namely 9.7813 mg/kg BW of rats, caused death and affected the brain and pancreas at doses of 27.3 mg/kg BW and 354.9 mg/kg BW which were characterized by edema and congestion. mild to moderate in the brain, and mild edema in the pancreas.
