Abstract

Penggunaan tonikum berbahan alam semakin digemari dikarenakan meningkatnya pola aktivitas masyarakat. Tonikum dapat menambah tenaga pada tubuh. Cabe jawa dan madu randu merupakan contoh tonikum berbahan alam mengandung alkaloid. Riset ini bertujuan guna mengetahui adanya pengaruh efek tonikum dan dosis efektifnya dari kombinasi ekstrak buah cabe jawa juga madu randu pada mencit Swiss Webster. Pengujian efek tonikum menggunakan metode natatory exhaustion dan uji gelantung. 7 kelompok perlakuan dibentuk dari 35 hewan uji. Kelompok I kontrol positif kafein 100 mg/KgBB. Kelompok II kontrol negatif CMC Na 0,5%. Kelompok III ekstrak cabe jawa tunggal 26 mg/KgBB. Kelompok IV madu randu tunggal 800 mg/KgBB. kelompok V kombinasi (25:75) ekstrak cabe jawa 6,5 mg/KgBB dan madu 600 mg/KgBB, kelompok VI kombinasi (50:50) ekstrak cabe jawa 13 mg/KgBB dan madu 400 mg/KgBB, kelompok VII kombinasi (75:25) ekstrak cabe jawa 19,5 mg/KgBB dan madu 200 mg/KgBB. Data efek tonikum berdasarkan dari selisih durasi mencit berenang (swimming time), penurunan %Immobility time, dan selisih durasi gelantung mencit. Analisis data menggunakan One-way Anova dilanjutkan dengan uji Post Hoc menggunakan Tukey. Kombinasi ekstrak cabe jawa dan madu randu terbukti mempunyai aktivitas tonikum atas mencit putih jantan galur Swiss Webster. Kombinasi ekstrak cabe jawa : madu (75 : 25) dengan dosis 19,5 mg/KgBB : 200 mg/KgBB menjadi kelompok teraktif dengan peningkatan durasi renang selama 443,4 detik, penurunan %Immobility time sebesar 25,36%, dan peningkatan durasi gelantung selama 73 detik. Kombinasi ini memiliki perbedaan hasil yang signifikan dengan dosis tunggal madu randu dan memiliki hasil yang lebih baik dibanding dosis tunggal cabe jawa. This research aims to determine tonic effects and effective doses from the combination of these two natural ingredients on Swiss Webster mice. Methods in research using natatory exhaustion and hangover test. 7 treatment groups were formed from 35 test animals. Group I positive control of caffeine 100 mg/KgBB. Group II negative control CMC Na 0.5%. Group III java long pepper extract 26 mg/KgBB. Group IV randu honey 800 mg/KgBB. group V combination (25:75) java long pepper extract 6.5 mg/KgBB : honey 600 mg/KgBB, group VI combination (50:50) java long pepper extract 13 mg/KgBB : honey 400 mg/KgBB, group VII combination (75:25) java long pepper extract 19.5 mg/KgBB : honey 200 mg/KgBB. Data tonic effect based on the difference duration of mice swimming (swimming time), decrease %Immobility time, and difference duration of mouse coils. Data analysis using One-way Anova followed by Post Hoc test using Tukey. The combination was shown to have tonic activity over Swiss Webster mice. The combination (75: 25) became the most active group with an increase in swimming duration for 443.4 seconds, a decrease in %Immobility time by 25.36%, and an increase in the duration of coil for 73 seconds. This combination has a significant difference in results with a single dose of randu honey and has better results than a single dose of java long pepper.
