Abstract
Pneumonia adalah infeksi radang parenkim yang disebabkan
oleh infeksi mikroorganisme, salah satunya bakteri Klebsiella
pneumoniae. Pemeriksaan sputum adalah salah satu langkah untuk
diagnosis pneumonia dengan mengetahui bakteri yang menginfeksi.
Daun mimba mengandung senyawa saponin, alkaloid, flavonoid,
triterpenoid, steroid, dan tanin yang berguna sebagai antibakteri.
Tujuan penelitian ini adalah untuk mengetahui ekstrak daun mimba
mampu menghambat bakteri Klebsiella pneumoniae pada isolat sputum
pasien penderita pneumonia dan berapa konsentrasi efektif daun mimba
sebagai antibakteri.
Ekstraksi daun mimba menggunakan metode maserasi dengan
pelarut etanol 96%. Isolat sputum diambil pada seorang pasien dengan
diagnosa pneumonia HAP yang memenuhi kriteria. Uji antibakteri
menggunakan 5 kelompok pengujian yaitu kelompok I (kontrol negatif)
dosis DMSO 10%, kelompok II (kontrol positif) dosis ciprofloxacin
500 mg, dan kelompok III, IV serta V kelompok uji antibakteri dengan
konsentrasi 30%, 36%, dan 42%. Parameter yang diamati adalah daya
hambat pertumbuhan bakteri dengan metode difusi cakram. Data yang
diperoleh kemudian dianalisis dengan SPSS menggunakan Kruskal
Wallis.
Hasil penelitian menunjukkan bahwa ekstrak daun mimba
mempunyai aktivitas antibakteri pada bakteri Klebsiella pneumoniae
dari isolate sputum pasien penderita pneumonia dengan konsentrasi
efektif 42%.
Pneumonia is an inflammation of the parenchyma caused by
microorganisms, one of which is the bacterium Klebsiella pneumoniae.
Sputum examination is one of the steps in diagnosing pneumonia by
knowing the infecting bacteria. Neem leaves contain saponins,
alkaloids, flavonoids, triterpenoids, steroids, and tannins that are useful
as antibacterial agents. The purpose of this study was to determine
whether neem leaf extract was able to inhibit Klebsiella pneumoniae
bacteria in the sputum isolates of patients with apneumonia.
Extraction of neem leaves using the gold maceration method
with 96% ethanol as a solvent Sputum isolates were collected from a
patients diagnosed with HAP pneumonia who met the criteria. The
antibacterial test used five groups, namely group kI (the negative
control) with a dose of 10% DMSO, group kII (the positive control)
with a dose of 500 mg ciprofloxacin, and groups III, IV, and V, the
antibacterial test groups, with concentrations of 30%, 36%, and 42%,
respectively. The parameter observed was the inhibition of bacterial
growth using the disc diffusion method. The data obtained was then
analyzed with SPSS using Kruskal Wallis.
The results showed that neem leaf extract had antibacterial
activity on Klebsiella pneumoniae isolates in the sputum of patients
with pneumonia, with an effective concentration of 42%.
