Abstract
Daya ingat merupakan kemampuan untuk mengingat dan
menyimpan informasi. Gejala terjadinya penurunan daya ingat adalah
demensia hingga kondisi terparah menyebabkan alzheimer. Penurunan
ini terjadi karena turunnya jumlah neurotransmiter asetilkolin dan
terjadi kerusakan sel piramidal di area hipokampus oleh beberapa
faktor. Gambir memiliki kandungan flavonoid dan fenolik yang
memiliki aktivitas dalam meningkatkan daya ingat. Tujuan penelitian
ini untuk mengetahui pemberian ekstrak daun gambir (EDG) dapat
meningkatkan daya ingat, dan mengetahui gambaran histopatologi
hipokampus pada otak mencit setelah diberikan ekstrak daun gambir.
Pengujian dilakukan pada 25 ekor mencit terbagi dalam 3
kelompok perlakuan yaitu kontrol postif (Ginkgo biloba), negatif (Na
CMC 1%), EDG dosis 500 mg/KgBB. Pengujian ini menggunakan
metode Morris Water Maze yaitu mencatat waktu latensi. Data waktu
latensi yang didapat dianalisis menggunakan uji Shapiro Wilk dan
Anova satu jalan.
Identifikasi senyawa dari serbuk daun gambir dan EDG
mengandung flavonoid, alkaloid, fenolik, tanin, steroid, triterpenoid
dan saponin. Hasil penelitian menunjukkan ekstrak daun gambir (EDG)
dosis 500 mg/KgBB memiliki aktivitas meningkatkan daya ingat. Dosis
500 mg/KgBB menunjukkan aktivitas peningkatan serta hasil gambaran
histopatologi hipokampus menunjukkan jumlah sel piramidal yang
paling baik mendekati Ginkgo biloba.
Memory is the ability to remember and retain information. The
symptom of memory decline can lead to dementia, and in severe cases,
it can cause Alzheimer's disease. This decline occurs due to a decrease
in the amount of acetylcholine neurotransmitter and damage to the
pyramidal cells in the hippocampus area caused by various factors.
Gambir (Uncaria gambir) contains flavonoids and phenolics that have
been shown to enhance memory. The aim of this study was to
investigate the effect of gambir leaf extract on memory enhancement
and to examine the histopathological changes in the hippocampus of
mice after administration of Gambir leaf extract.
The testing was conducted on 25 mice divided into three
treatment groups: positive control (Ginkgo biloba), negative control
(Na-CMC 1%), and GLE at a dose of 500 mg/kg body weight. The
Morris Water Maze method was used to record latency time. The
latency time data obtained were analyzed using the Shapiro-Wilk test
and one-way ANOVA.
The identification of compounds from Gambir leaf powder and
GLE revealed the presence of flavonoids, alkaloids, phenolics, tannins,
steroids, triterpenoids, and saponins. The research results showed that
gambir leaf extract at a dose of 500 mg/kg body weight exhibited
memory-enhancing activity. The 500 mg/kg body weight dose showed
the
most significant improvement, and the histopathological
examination of the hippocampus indicated that the number of
pyramidal cells approached the positive control (Ginkgo biloba) group,
suggesting a positive effect on memory enhancement.
