Abstract

Enzim fibrinolitik merupakan kelompok protease serin memiliki kemampuan untuk menghancurkan bekuan darah dalam berbagai jenis penyakit thrombosis seperti infark miokard akut. Mekanisme kerja enzim fibrinolitik dengan menghidrolisis fibrin yang menyebabkan bekuan darah menjadi produk terlarut yang dapat dibuang dari peredaran darah sehingga membebaskan pembuluh darah dari bekuan darah lalu memulai proses penyembuhan dinding pembuluh darah. Pemanfaatan bakteri sebagai penghasil enzim fibrinolitik dapat berasal dari limbah. Limbah cair industri kecap memiliki kandungan protein sehingga dapat digunakan sebagai sumber isolat bakteri fibrinolitik. Tujuan penelitian ini untuk mengetahui aktivitas fibrinolitik isolat bakteri limbah cair industri kecap. Penelitian ini menggunakan isolat bakteri dari limbah cair industri kecap. Metode diawali dengan pengambilan sampel, isolasi bakteri, identifikasi bakteri, pembuatan ekstrak kasar enzim, pemurniaan ekstrak kasar enzim, pembuatan sampel uji dan penentuan potensi fibrinolitik menggunakan metode clot lysis. Hasil persentase (%) lisis bekuan darah.yang dihasilkan menunjukkan kemampuan ekstrak enzim dalam mendegradasi fibrin. Analisis data menggunakan cara pendekatan statistik two way ANOVA. Hasil uji proteolitik bakteri yang diisolasi dari limbah cair industri kecap menghasilkan 4 koloni yang memiliki aktivitas proteolitik. Hasil identifikasi pewarnaan Gram menunjukan 3 bakteri Gram positif dan 1 bakteri Gram negatif. Ekstrak kasar enzim fibrinolitik yang berasal dari bakteri limbah cair industri kecap mampu melisiskan gumpalan darah. Konsentrasi optimal ekstrak kasar enzim fibrinolitik sebagai agen fibrinolitik secara in vitro pada setiap koloni yaitu konsentrasi 80%. Fibrinolytic enzymes are a group of serine proteases that have the ability to destroy blood clots in various types of thrombosis diseases such as acute myocardial infarction. The mechanism of action of fibrinolytic enzymes is by hydrolyzing fibrin that causes blood clots into soluble products that can be removed from the blood circulation, thus freeing blood vessels from blood clots and then starting the healing process of the blood vessel wall. The utilization of bacteria as fibrinolytic enzyme producers can be derived from waste. Soy sauce industry liquid waste has protein content so that it can be used as a source of fibrinolytic bacterial isolates. The purpose of this study was to determine the fibrinolytic activity of bacterial isolates of soy sauce industry liquid waste. This study used bacterial isolates from soy sauce industry liquid waste. The method begins with sampling, isolation of bacteria, identification of bacteria, preparation of enzyme crude extract, purification of enzyme crude extract, preparation of test samples and determination of fibrinolytic potential using the clot lysis method. The results of the percentage (%) of blood clot lysis produced indicate the ability of the enzyme extract to degrade fibrin. Data analysis using two way ANOVA statistical approach. The results of the proteolytic test of bacteria isolated from soy sauce industry liquid waste produced 4 colonies that had proteolytic activity. Gram stain identification results showed 3 Gram positive bacteria and 1 Gram negative bacteria. Crude extract of fibrinolytic enzyme derived from soy sauce industry liquid waste bacteria is able to lyse blood clots. The optimal concentration of fibrinolytic enzyme crude extract as a fibrinolytic agent in vitro on each colony is 80% concentration.
