Abstract

Negara Cina, Korea, dan Vietnam dalam pengobatan tradisional menggunakan Lumbricus rubellus untuk menyembuhkan berbagai penyakit infeksi saluran pencernaan, infeksi saluran pernapasan, substrat pelembut kulit, pelembab wajah, dan anti infeksi. Kandungan kimia Lumbricus rubellus yang mempunyai aktivitas antibakteri adalah lumbricin-1. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri ekstrak Lumbricus rubellus dengan pelarut asam asetat berdasarkan konsentrasi hambat minimum (KHM) berdasarkan metode difusi dan mengetahui perbandingan aktivitas antibakteri terhadap Bacillus cereus dan Klebsiella pneumoniae. Penelitian eksperimental menggunakan cacing tanah segar sebanyak 16 kg dibersihkan, dicuci, ditiriskan, dikeringkan di bawah sinar matahari hingga kering, dihancurkan hingga menjadi serbuk. Serbuk cacing tanah diekstraksi menggunakan metode maserasi dengan pelarut asam asetat 20%; 40%; 80% yang selanjutnya diuji aktivitas antibakteri menggunakan metode difusi dengan konsentrasi setiap ekstrak 50% untuk mengetahui aktivitas antibakteri antara Bacillus cereus ATCC 10876 dan Klebsiella pneumoniae ATCC 70063 menggunakan ekstrak cacing tanah serta untuk mengetahui ekstrak cacing tanah manakah yang teraktif dalam menghambat Bacillus cereus ATCC 10876 dan Klebsiella pneumoniae ATCC 70063 dan dilakukan analisis data dengan spss menggunakan metode analisis oneway anova dan T test. Ketiga ekstrak cacing tanah yang dimaserasi dengan pelarut asam asetat terbukti mempunyai aktivitas antibakteri terhadap Bacillus cereus ATCC 10876 dan Klebsiella pneumoniae ATCC 70063. Ekstrak cacing tanah yang dimaserasi asam asetat 80% mempunyai aktivitas antibakteri teraktif dibandingkan dengan ekstrak yang dimaserasi pelarut asam asetat 40% dan 20% yang dibuktikan dengan uji difusi. Ekstrak cacing tanah memiliki aktivitas antibakteri yang baik dalam menghambat Bacillus cereus ATCC 10876 dibandingkan Klebsiella pneumoniae ATCC 70063. Chinese, Korean and Vietnamese countries in traditional medicine use Lumbricus rubellus to cure various infectious diseases of the digestive tract, respiratory tract infections, skin softening substrates, facial moisturizers, and anti-infectives. The chemical substance of Lumbricus rubellus which has antibacterial activity is lumbricin-1. The purpose of this study was to determine the antibacterial activity of Lumbricus rubellus extract with acetic acid solvent based on the minimum inhibitory concentration (MIC) based on the diffusion method and to determine the comparison of antibacterial activity against Bacillus cereus and Klebsiella pneumoniae. Experimental research used 16 kg of fresh earthworms cleaned, washed, drained, dried in the sun to dry, crushed to a powder. Earthworm powder was extracted with 20% acetic acid solvent; 40%; 80%, the extracts obtained were then tested for antibacterial activity using the diffusion method with each extract of earthworms being made into an extract test preparation of 50% to determine the most active extract of the three extracts of earthworms which were then analyzed using SPSS.
