Abstract
Rimpang lengkuas (Alpinia galanga (L.) Sw.) merupakan salah
satu bahan baku tanaman obat tradisional. Bahan baku obat tradisional
yang baik harus memenuhui syarat mutu, aman dan berefikasi. Lokasi
geografis tanam menjadi salah satu faktor eksternal yang dapat
mempengaruhi mutu simplisia dan ekstrak. Penelitian ini bertujuan
untuk mengetahui apakah perbedaan ketinggian lokasi tanam
berpengaruh terhadap kualitas dan profil metabolit sekunder pada
rimpang lengkuas.
Sampel rimpang lengkuas diperoleh dari Kecamatan Jaten yang
berada di ketinggian geografis di dataran rendah dengan rata-rata
ketinggian 90 mdpl, dataran sedang di Jumantono 450 mdpl dan
dataran tinggi di Jenawi 1000 mdpl. Pengujian kualitas bahan baku
dilakukan melalui parameter non spesifik secara fisika dan spesifik
(profil metabolit sekunder yang diidentifikasi menggunakan metode
HPTLC menggunakan pembanding eugenol). Parameter uji spesifik
non spesifik dianalisa secara SPSS Pearson Correlation. Profil
metabolit dianalisis menggunakan principal component analysis (PCA)
dan hierarchical clustering analysis (HCA).
Terdapat pengaruh ketinggian lokasi tanam terhadap nilai kadar
sari larut air dan etanol sebagai parameter uji spesifik pada lengkuas
dengan nilai 2-tailed <0,05 masing-masing 0,014 dan 0,011 dengan
nilai dengan kategori kuat dengan nilai Correlation Coefficient -.716*
dan-.738*. namun tidak mempengaruhi pada parameter uji non spesifik.
Berdasarkan hasil analisis PCA tedapat keragaman pada profil
metabolit sekunder dengan nilai eigenvalue 8,0703; 6,6445; 2,3558;
2,0467; 1,6452; 1,3221 pada PC1 sampai PC6. Sampel jenawi memiliki
perbedaan dengan sampel Jaten dan Jumantono dengan nilai similarity
61,49% dengan analisis HCA.
Galangal rhizome (Alpinia galanga (L.) Sw.) is one of the raw
materials for medicinal plants that must be of good quality in order to
increase its quality, safety and benefits. The geographical location of
planting is one of the external factors that can affect the quality of the
simplicia, extracts and secondary metabolite content produced. This
study aims to determine whether the difference in the height of the
planting location affects the quality and profile of secondary
metabolites in galangal (Alpinia galanga (L.) Sw.) rhizomes.
Samples were obtained from Jaten District as a location with
geographical elevation in the lowlands with an average altitude of 90
masl, Jumantono plains 450 masl and Jenawi highlands 1000 masl.
The quality of raw materials is tested through non-specific
physical and specific parameters (secondary metabolite profiles
identified using the HPTLC method using eugenol as a comparison).
Specific and non-specific parameters were analyzed using SPSS
Pearson Correlation. Metabolite profiles were analyzed using principal
component analysis (PCA) and hierarchical clustering analysis (HCA).
The results of statistical tests with Pearson Correlation show
that there is an influence of the height of the planting location on the
value of water soluble content and ethanol content as specific test
parameters for galangal with 2-tailed values < 0.05 respectively of
0.014 and 0.011 with values in the strong category with Correlation
Coefficient values of -.716* and -.738*, but did not affect non-specific
test parameters. Based on the results of PCA analysis, there is a
diversity of secondary metabolite profiles with an eigenvalue of 8.0703;
6.6445; 2.3558; 2.0467; 1.6452; 1.3221 on PC1 to PC6. The Jenawi
sample is different from the Jaten and Jumantono samples with a
similarity value of 61.49% using HCA analysis.
