Abstract
Depresi merupakan gangguan yang ditunjukkan dengan adanya
perubahan suasana mood, pola aktivitas seseorang yang berbeda
daripada biasanya. Depresi dialami oleh remaja hingga usia tua,
sehingga untuk mengatasi depresi masyarakat dapat mengkonsumsi
obat dari bahan herbal. Salah satunya adalah daun singkong yang
memiliki khasiat sebagai antidepresan. Penelitian ini bertujuan untuk
mengetahui efek antidepresan dari ekstrak daun singkong yang
diberikan secara oral pada mencit putih jantan.
Pengujian efek antidepresan ini dilakukan dengan parameter
yaitu %immobility time dengan pengujian forced swimming test.
Sehingga mencit dibagi menjadi 5 kelompok yang terdiri dari 5 mencit
ditiap kelompok. Kelompok 1 diberi kontrol positif (amitriptilin),
kelompok 2 diberi kontrol negatif (CMC Na 0,5%), kelompok 3, 4, 5
diberi ekstrak etanol daun singkong dengan konsentrasi 125, 250, 500
mg/KgBb.
Daun singkong mengandung flavonoid spesifik golongan rutin
yang bermanfaat sebagai antidepresan. Dosis ekstrak etanol daun
singkong 125 mg/Kg %immobility time sebesar 28,76%. Dosis ekstrak
etanol daun singkong 250 mg/KgBb dengan % immobility time 35,14%.
Dosis efektif ekstrak etanol daun singkong dengan dosis 500mg/KgBb
karena memiliki efek antidepresan yang setara dengan kontrol positif
amitripilin dengan %immobility time sebesar 44,94%.
Depression is a disorder indicated by a change in mood, a
person's pattern of activity that is different than usual. Depression is
experienced by teenagers to old age, so to overcome depression, people
can consume drugs made from herbal ingredients. One of them is
cassava leaves which have properties as an antidepressant. This study
aims to determine the antidepressant effect of cassava leaf extract given
orally to male white mice.
Testing the effect of this antidepressant was carried out by %
immobility time with the forced swimming test. So the mice were
divided into 5 groups consisting of 5 mice in each group. Group 1 was
given a positive control (amitriptyline), group 2 was given a negative
control (CMC Na 0.5%), groups 3, 4, 5 were given ethanol extract of
cassava leaves with concentrations of 125, 250, 500 mg/KgBb.
Cassava leaves contain specific flavonoids of the rutin group
which are useful as antidepressants. The dose of cassava leaf ethanol
extract of 125 mg/Kg % immobility time of 28.76%. The dose of
ethanol extract of cassava leaves 250 mg/KgBb has a % immobility
time of 35.14%. The effective dose of cassava leaf ethanol extract at a
dose of 500 mg/Kg Bb because it has an antidepressant effect that is
equivalent to the positive control of amitriptyline with a % immobility
time of 44.94%.
