Abstract

Rimpang lengkuas merah (Alpinia purpurata K. Schum.) memiliki kandungan flavonoid, tanin, alkaloid, dan triterpenoid yang dapat digunakan sebagai antibakteri. Sediaan emulgel merupakan sediaan topikal yang menguntungkan terutama pada sistem penghantarannya. Emulgel memiliki keuntungan penetrasi yang baik, dengan dosis minimal mampu menimbulkan efek farmakologis. Tujuan penelitian ini untuk memformulasikan sediaan emulgel ekstrak rimpang lengkuas merah dengan variasi konsentrasi HPMC untuk menghasilkan mutu fisik, stabilitas yang baik, serta mempunyai aktivitas antibakteri terhadap Staphylococcus aureus dan untuk mengetahui formula yang paling baik yang diperoleh. Metode ekstraksi yang digunakan adalah maserasi dengan pelarut etanol 96%. Ekstrak rimpang lengkuas merah diformulasikan menjadi sediaan emulgel, dengan variasi HPMC sebesar 2%, 3%, dan 4%. Sediaan emulgel dievaluasi mutu fisik dan stabilitas, kemudian dilanjutkan uji aktivitas antibakteri menggunakan metode difusi sumuran. Data yang didapat kemudian dianalisis statistik menggunakan aplikasi SPSS 25. Peningkatan konsentrasi HPMC menyebabkan peningkatan viskositas, daya lekat dan pH, namun mengalami penurunan daya sebar dan nilai daya hambat bakteri. Semua formula 1, 2, dan 3 memiliki aktivitas antibakteri. Formula 1 sediaan emulgel ekstrak rimpang lengkuas merah memiliki mutu fisik yang paling baik dengan nilai daya hambat yang tergolong kuat yaitu 15,52±0,15 mm. Semua formula memiliki stabilitas yang tidak stabil pada parameter tertentu pada pengujian sesudah cycling test. Red galangal rhizome (Alpinia purpurata K. Schum.) contains flavonoids, tannins, alkaloids, and triterpenoids, which can be used as antibacterials. Emulgel preparations are topical preparations with advantages, especially in their delivery system. Emulgels have the advantage of good penetration, and even minimal doses can produce pharmacological effects. The purpose of this study was to formulate emulgel preparations from red galangal rhizome extract with varying HPMC concentrations to produce good physical quality and stability and antibacterial activity against Staphylococcus aureus. The extraction method used was maceration with 96% ethanol. Red galangal rhizome extract was formulated into emulgel preparations, with varying HPMC concentrations of 2%, 3%, and 4%. The emulgel preparations were evaluated for physical quality and stability, followed by antibacterial activity testing using the well diffusion method. The data obtained were then statistically analyzed using SPSS 25. Increasing the HPMC concentration resulted in increased viscosity, adhesiveness, and pH, but decreased spreadability and bacterial inhibition values. All formulas 1, 2, and 3 exhibited antibacterial activity. Formula 1, a red galangal rhizome extract emulgel preparation, had the best physical quality, with a relatively strong inhibition value of 15,52±0,15 mm. All formulas exhibited unstable stability in certain parameters after cycling
