Abstract
Obat merupakan zat yang dapat mempengaruhi senyawa di
dalam tubuh untuk mencegah, mengobati, atau mendiagnosis suatu
penyakit, serta menimbulkan kondisi tertentu. Obat tersedia dalam
berbagai bentuk sediaan, salah satunya adalah sediaan oral yang paling
umum digunakan karena praktis, meskipun memiliki kelemahan berupa
penyerapan yang lambat sehingga waktu mula kerja menjadi lebih
lama. Paracetamol yang umumnya dikonsumsi dalam bentuk tablet
dapat dimodifikasi menjadi tablet effervescent yang lebih praktis dan
cepat larut. Perbedaan konsentrasi bahan pengikat polyvinylpyrrolidone
(PVP) berpotensi mempengaruhi karakteristik fisik granul dan tablet
effervescent.
Penelitian ini bertujuan untuk mengetahui pengaruh variasi
konsentrasi PVP terhadap sifat fisik tablet effervescent paracetamol.
Pembuatan tablet dilakukan dengan metode granulasi basah
menggunakan tiga variasi konsentrasi PVP, yaitu 0,01%, 0,05%, dan
0,1%. Pengujian yang dilakukan meliputi keseragaman bobot,
kekerasan, kerapuhan, waktu larut, serta uji kesukaan rasa oleh
responden.
Hasil penelitian menunjukkan bahwa seluruh formula
memenuhi persyaratan keseragaman bobot. Peningkatan konsentrasi
PVP berbanding lurus dengan nilai kekerasan tablet, di mana PVP
0,1% memiliki kekerasan tertinggi sebesar 4,5 kgf, sekaligus
menghasilkan kerapuhan terendah sebesar 0,21%. Sebaliknya, waktu
larut meningkat dengan bertambahnya konsentrasi PVP; formula 0,01%
memiliki waktu larut tercepat selama 1 menit 45 detik, sedangkan
formula 0,1% terlama selama 3 menit 12 detik. Uji kesukaan rasa
menunjukkan bahwa formula dengan PVP 0,05% memperoleh skor
tertinggi, sehingga dinyatakan memiliki kombinasi terbaik antara sifat
fisik dan penerimaan rasa.
A drug is a substance capable of affecting compounds within
the body to prevent, treat, or diagnose a disease, as well as to induce
certain physiological conditions. Drugs are available in various dosage
forms, one of which is the oral dosage form, the most commonly used
due to its practicality, although it has the drawback of slow absorption,
resulting in a longer onset of action. Paracetamol, which is commonly
consumed in tablet form, can be modified into an effervescent tablet
that is more practical and dissolves more rapidly. Variations in the
concentration of the binder polyvinylpyrrolidone (PVP) may influence
the physical characteristics of granules and effervescent tablets.
This study aimed to determine the effect of varying PVP
concentrations on the physical properties of paracetamol effervescent
tablets. The tablets were prepared using the wet granulation method
with three different PVP concentrations: 0.01%, 0.05%, and 0.1%. The
evaluations conducted included weight uniformity, hardness, friability,
dissolution time, and taste preference assessment by respondens.
The results showed that all formulations met the requirements
for weight uniformity. Increasing PVP concentration correlated
positively with tablet hardness, where 0.1% PVP produced the highest
hardness value of 4.5 kgf and the lowest friability of 0.21%.
Conversely, dissolution time increased with higher PVP concentrations;
the 0.01% formulation had the fastest dissolution time of 1 minute 45
seconds, whereas the 0.1% formulation had the slowest at 3 minutes 12
seconds. Taste preference testing indicated that the 0.05% PVP
formulation received the highest score, suggesting it provided the best
combination of physical properties and taste acceptability.
