Abstract
Daun pepaya (Carica papaya L.) mengandung metabolit
sekunder flavonoid, alkaloid, tanin dan saponin yang diketahui terbukti
berpotensi sebagai antidepresan. Flavanoid ditemukan sebagai senyawa
utama pada daun pepaya. Flavanoid diketahui terbukti berpotensi
sebagai antidepresan. Tujuan dari penelitian ini adalah untuk
mengetahui aktivitas antidepresan dari ekstrak etanol daun pepaya
(Carica papaya L.) pada mencit putih jantan (Mus Muculus) dengan
metode FST (Forced Swim Test) dan mengetahui dosis efektif ekstrak
etanol daun pepaya sebagai antidepresan.
Penyarian ekstrak daun papaya menggunakan metode maserasi
dengan etanol 96% sebagai pelarut. Penelitian ini menggunakan 30
mencit putih jantan berumur 2-3 bulan dengan berat badan 25-33 g
yang dibagi menjadi 6 kelompok, terdiri dari kontrol positif
(Amitriptilyn), kontrol negatif (CMC Na 0,5%), ekstrak daun pepaya
100, 200, dan 400 mg/kgBB serta kontrol tanpa perlakuan. Kelompok
satu sampai lima diberi perlakuan yang sama, kelompok enam tanpa
perlakuan induksi stress. Selama 7 hari mencit yang depresi diinduksi
dengan metode Tail Suspension Test dan suspensi oral diberikan selama
7 hari berikutnya. Pada hari ke 14 waktu imobilitas diukur pada mencit.
Data dari pengukuran imobilitas dianalisis menggunakan One Way
Anova untuk menentukan normalitas dan homogenitas data kemudian
dilanjutkan dengan uji tukey HSD (Honestly Significant Different).
Hasil penelitian yang telah dilakukan menunjukkan bahwa
pemberian ekstrak daun papaya 100, 200, dan 400 mg/kgBB dapat
mengurangi immobility time dengan cara menghambat aktivitas
monoamine oksidase MAO-A. Dosis paling efektif adalah 400 mg/
kgBB sebagai antidepresan.
Papaya leaves (Carica papaya L.) contain secondary metabolites
such as flavonoids, tannins, and alkaloids known to have the potential
as antidepressants. The aim of this study was to determine the
antidepressant activity of papaya leaf ethanol extract (Carica papaya L.)
on male white mice (Mus Muculus) using the Forced Swim Test (FST)
method, as well as to identify the effective dosage of papaya leaf
ethanol extract as an antidepressant.
The papaya leaf extract was obtained using the maceration
method with 96% ethanol as the solvent. The study involved 30 male
white mice, aged 2-3 months, weighing 25-33 g, divided into 6 groups.
These groups consisted of a positive control (Amitriptyline), negative
control (0.5% CMC Na), and papaya leaf extract at doses of 100, 200,
and 400 mg/kg body weight, as well as a control group without any
treatment. Groups one to five received the same treatment, while group
six remained untreated for stress induction. For 7 days, the mice in a
state of depression were induced using the forced swim test and the
Tail Suspension Test, followed by oral suspension administration for
the next 7 days. On the 14th day, the immobility time was measured in
the mice. The data obtained from the immobility measurement were
analyzed using One-Way ANOVA to determine data normality and
homogeneity, followed by the Tukey HSD (Honestly Significant
Difference) test.
The results of the study showed that the administration of
papaya leaf extract could reduce immobility time by inhibiting the
activity of monoamine oxidase MAO-A. The most effective dosage
was found to be 400 mg/kg body weight as an antidepressant.
